Dominant mutation for nystatin resistance in yeast.

In general, attempts to isolate strains of Candida albicans resistant to the commonly employed antifungicide nystatin have been unsuccessful (M. L. Littman, M. A. Pisano, and R. M. Lancaser, Antibiotics Ann. 1957-58, p. 981, 1958; C. K. J. Paniker, P. V. Kurup, and K. Indira Devi, Indian J. Med. Res. 51:846, 1963). A few strains, however, have yielded variants exhibiting threeto fourfold resist...

Self-induced nystatin resistance in Dictyostelium discoideum.

The cellular slime mold Dictyostelium discoideum is normally susceptible to the action of the polyene antibiotic nystatin. We describe conditions under which transient nystatin resistance can be induced in the absence of detectable changes in sterol composition. These results suggest that nystatin plays a positive role in promoting resistance to itself.

Nystatin-resistant mutants of yeast: alterations in sterol content.

Mutants of the genes nys1 and nys3 differ from sensitive strains (nys(+)) in their sterol content. Ultraviolet absorption spectra of the nonsaponifiable material extracted from cells of nys(+) demonstrated the presence of ergosterol and 24(28)-dehydroergosterol. In nys1 mutants, the spectrum suggests the presence of a new sterol. The absorption spectrum of extracts from nys3 mutants indicates a...

Dominant chromosomal mutation bypassing chromosomal genes needed for killer RNA plasmid replication in yeast.

Yeast strains carrying a double-stranded RNA plasmid of 1.4-1.7 x 10(6) daltons encapsulated in virus-like particles secrete a toxin that kills strains lacking this plasmid. The plasmid requires at least 24 chromosomal genes (pets, and mak1 through mak23) for its replication or maintenance. We have detected dominant Mendelian mutations (called KRB1 for killer replication bypass) that bypass two...
